INTRODUCTION
Japanese encephalitis virus (JEV) is a flavivirus related to dengue, yellow fever and West Nile viruses, and is spread by mosquitoes.JEV is the main cause of viral encephalitis in m any countries of Asia with an estimated 68 000 clinical cases every year.24 countries in th e WHO South-East Asia and Western Pacific regions have endemic JEV transmission, ex posing more than 3 billion people to risks of infection.
TEST PRINCIPLE
Mytest JEV IgM Rapid Test is a lateral flow chromatographic immunoassay based on the principle of the IgM capture assay. The test cassette consists of: 1) a conjugate pad containing Anti-Human IgM antibody conjugated with colloid gold 2) a nitrocellulose membrane strip containing a test band (T band) and a control band (C band). The T band is pre-coated with Rec. JEV Antigen, and the C band is pre-coated with goat anti-mouse IgG. When an adequate volume of test specimen is dispensed into device, it migrates by capillary action across the cassette. The IgM antibody to JEV, if present in the specimen will bind to the Anti-Human IgM conjugates. The immuno complex is then captured on the membrane by the pre-coated Rec. JEV Ag, forming a colored T band, indicating a JEV IgM reactive test result. Absence of the T band suggests a non-reactive result. The test contains an internal control (C band) which should exhibit a colored band of the immuno complex of Mouse Antibody+Goat Anti-mouse regardless of the color development on the T band. Otherwise, the test result is invalid and the specimen must be retested with another device.
INTENDED USE
JEV Test is a rapid immunochromatography assay for the detection of IgM antibodies to JE virus in human whole blood, serum or plasma. The assay is used as a screening test for JEV infection and as an aid for diagnosis of infections in conjunction with other criteria.
