Oscar HIV (4th Generation ELISA) detects antigen and antibody of HIV-1 & 2 in human serum / plasma. It is available in 96T pack. Microwell strips are coated with monoclonal p24 antibodies along with recombinant antigens gp 120, gp41 and gp36 representing both HIV-1 and HIV-2. Samples, positive and negative controls are added along with biotinylated p24 antibody in the coated wells and incubated.
The wells are washed to remove unbound components and HIV1 and HIV2 recombinant antigens conjugated to HRP along with streptavidin conjugated to HRP are added. After a short incubation the wells are washed again and bound enzyme is detected by adding substrate. The reaction is stoped after specified time with acid and absorbance is determined for each well at 450 nm with an ELISA reader. The cut-off value is calculated by the given formula and absorbance of all the wells are compared with the cut-off value. Any sample having absorbance more than the cut-off value is considered reactive.

INTRODUCTION
Q-Fever is a disease that results from infection with small, polymorph and gram-negative bacteria called Coxiella burnetii. After an outbreak in Brisbane, Australia, the responsible organism was isolated and named Coxiella burnetii in honour of Dr. Herald Rae Cox and Sir Frank Burnet. New molecular research demonstrated a close relationship to Legionella. The zoonosis Q-Fever is found everywhere except New Zealand (no data available). There is an extensive reservoir (mainly ticks) of C. burnetii. Ticks are an important vector of the pathogen in the transmission between domestic and wildlife animals. But the ticks are unimportant in the direct infection of humans. Cattles, sheep and goats are usually the source of transmission of this microorganism to humans. However cats, dogs and rabbits are also important in this regard. In most instances humans become infected with Coxiella burnetii following inhalation of contaminated aerosols (respiratory tract). The incubation period for Q-Fever in humans is about 2 weeks. The resulting illness can be divided into acute and chronic varieties. During the acute phase of illness antibodies to the phase II-antigen are formed. Anti phase-I antibodies in high titers are typical for a chronic disease. In areas where Q-Fever is endemic, 12% or more of the population have antibodies to C. burnetii. Most of the infections are subclinical or undiagnosed.

The JE Detect™ IgM Antibody Capture ELISA (MAC-ELISA) test for exposure to Japanese Encephalitis Virus (JEV) is an ELISA assay system for the detection of IgM antibodies in human serum to JEV-derived recombinant antigen (JERA) (1-4). SUMMARY AND EXPLANATION OF THE TEST -
Exposure to JEV causes a disease with a number of symptoms including encephalitis (5-8). JE Detect™ IgM Antibody Capture ELISA (MAC-ELISA) employs a recombinant antigen called JERA, which can be used as a rapid serological marker for JEV infection. The JERA protein is a recombinant antigen, which consists of a stretch of peptides from different parts of the JEV.
PRINCIPLE OF THE TEST
The JE Detect™ IgM Antibody Capture ELISA (MAC-ELISA) consists of one enzymatically-amplified "two-step" sandwichtype immunoassay. In this assay, JE Detect™ Negative Control (represents non-reactive serum), JE Detect™ IgM Positive Control (represents reactive serum), and unknown serum samples are diluted with Sample Dilution Buffer, then incubated in microtitration wells which have been coated with anti-human IgM antibodies. This is followed by incubation with both JEVderived recombinant antigen (JERA) and Normal Cell Antigen (NCA) separately. After incubation and washing, the wells are treated with a JERA-specific antibody labeled with the enzyme horseradish peroxidase (HRP). After a third incubation and washing step, the wells are incubated with the tetramethylbenzidine (TMB) substrate. An acidic stopping solution is then added and the degree of enzymatic turnover of the substrate is determined by absorbance measurement at 450 nanometers. Above a certain threshold, the ratio of the absorbencies of the JERA and the control wells accurately determines whether antibodies to JEV are present

INTRODUCTION
Autoimmunity is the failure of an organism to recognize its own constituent parts as self, which allows an immune response against its own cells and tissues. Any disease that results from such an aberrant immune response is termed an Autoimmune Disease. Rheumatoid autoimmune diseases are often associated with the occurrence of autoantibodies against several nuclear or cytoplasmatic antigens. We can to distinguish between Anti Nuclear Antibodies (ANA), associate with autoimmune systemic diseases as SLE (Systemic Lupus Erythematosus), RA (Reumatoid Arthritis), Scleroderma, MCDT (Mixed Connective Tissue Disease) and Sjogren’s Syndrome; and Extractable anti Nuclear Antibodies (ENA), associate with Polymyositis, SLE, MCDT and Sjogren’s Syndrome. Ro/SSA and La/SSB are cytoplasmatic antigens that occur in combination in patients with Sjögren-Syndrome. Autoantibodies against SSA-60 KDa ribonucleoprotein are clinically important since they are associated with subacute cutaneous lupus erythematosus (SCLE) and neonatal lupus erythematosus (NLE). Anti-Ro/SSA are present in approximately 70% of such patients.
PRINCIPLE OF THE TEST
Microplates are coated with a preparation of recombinant SSA 60KDa antigen. In the 1st incubation, the solid phase is treated with diluted samples and anti-SSA60 IgG are captured, if present, by the solid phase. After washing out all the other components of the sample, in the 2nd incubation bound anti-SSA60 are detected by the addition of anti hIgG antibody, labelled with peroxidase (HRP). The enzyme captured on the solid phase, acting on the substrate/chromogen mixture, generates an optical signal that is proportional to the amount of anti-SSA60 IgG antibodies present in the sample. IgG in the sample may be quantitated by means of a standard curve calibrated in arbitrary units per milliliter (arbU/ml) as no international standard is available.

IgE constitutes a fraction of the total antibody in serum 50-300 ng/ml (compared to 10 mg/ml for IgG) and together with its Fc receptor is important in primary immune responses. The immunogenetic mechanisms underlying IgE responsiveness seen in the atopic diseases can be divided into antigen-specific and non-antigen-specific responses. Parasitic infestations such as hookworm, and certain clinical disorders including aspergillosis, have also been demonstrated to cause high levels of IgE. Decreased levels of IgE are found in cases of hypogammaglobulinemia, autoimmune diseases, ulcerative colitis, hepatitis, cancer, and malaria. Cord blood or serum IgE levels may have prognostic value in assessing the risk of future allergic conditions in children. Certain groups of white blood cells, including basophils and tissue mast cells, have membrane receptors for the IgE molecule.

This test kit is intended for use in the quantitative determination of total 25-OH Vitamin D (Vitamin D2 and Vitamin D3) in human serum and plasma. This kit is for in vitro diagnostic use only
PHYSIOLOGY -
The group of compounds referred to as Vitamin D, are actually fat soluble steroidal prehormones. The main forms which occur in the body are Vitamin D2 (ergocalciferol) and Vitamin D3 (cholecalciferol). The active form of these molecules is Dihydroxyvitamin D3 (1, 25(OH)2 D3). Vitamin D3 is formed in the skin by photolysis of 7-dehydrocholesterol by ultraviolet radiation from sunlight. It is transported in blood circulation bound to proteins to the liver where it is hydroxylated. Further hydroxylation occurs in the kidneys to produce the most active form. Vitamin D levels are highest in newborns and decrease exponentially throughout life. Sufficient circulating levels of vitamin D are necessary for healthy bone maintenance and cell metabolism. Recent studies have shown that it may also lower incidents of certain cancers. Insufficient levels of Vitamin D can result in osteoporosis and bone fracture in the elderly, secondary hyperparathyroidism, abnormal cell metabolism and even increased incidents of cancer. Severe deficiency may lead to rickets in children and osteomalacia in adults. Disease associated with Vitamin D deficiency may also include: impaired immunity, increased autoimmunity, myopathy, diabetes mellitus, and an increased risk of colon, breast, and prostate cancers. Abnormally high levels (> 200 ng/ml) of Vitamin D leads to Vitamin D toxicity and may cause hypercalcaemia.
